3D Cell Culture Basics: Forming Spheroids in the Presence of Extracellular Matrix

Abstract

The PowerPoint file combined with the accompanying videos comprises the introductory training presentation for 3D Cell Culture Basics that make use of gels and is focused on techniques to be used for cells that form spheroid-like structures in vivo, notably a number of tumors and certain normal epithelial and neural structures.

The purpose of 3D cell culture is to place cells in an environment in which they reproduce physiologically relevant features of tissues. There is a wide range of techniques in setting up 3D cell cultures; however, fundamentally, the aim is to help cells behave as they would in the body by bringing, most often simple, modifications of the cell culture environment. The presentation covers some basic techniques for the setup of cell cultures using drip and thick embedding methods with EHS (Engelbreth-Holm-Swarm)-derived gel. Note that for each type of gel used, it is important to follow the manufacturer’s instructions and publications if available. Our presentation highlights fundamental principles of cell culture with a gel. The presentation also covers adding and removing cell culture medium in the presence of any gel, the use of serum-free medium, dislodging multicellular structures from Matrigel-based 3D culture and preparing 3D cultures for immunolabeling.
